pH Dependent Preparation of Hyaluronic Acid Decorated Chitosan Nanoparticles for drug delivery applications

Abstract:
The purpose of this study was to produce nanoparticles made from chitosan (CS) and Hyaluronic acid (HA) in solutions of different pH of CS and different HA/CS volume ratio for biomedical drug delivery applications. Ionic gelation technique was used followed to make the nanoparticles via using triphosphate (TPP) as crosslinker and then in order to characterize nanoparticles, morphology of nanoparticles, HA loading efficacy and HA release from the nanoparticles were studied. Morphological characterization of the obtained nanoparticles using SEM showed that at pH=4.09 nanoparticles of uniform structure with almost spherical shape and narrow size distribution were obtained. The particle size was determined by Image j software and the average size of nanoparticles was 520±18nm. Studying of HA loading efficacy and HA release in PBS showed that 97.66±2.0% of initial HA loaded on nanoparticles and for all the loaded HA, almost 67% of the initial HA released in first 2h and the remaining sustainably released within 3 days.
